It's impossible to assess such a thing remotely, so if you think you have a problem that needs looking at, please do take the car to a good mechanic.

Glass roofs can indeed allow more noise into the cabin, though smoothly integrated roof rails aren't usually an issue. Is this a new problem on a car you've had for a while? If so, then it's more likely to be something wrong in the suspension if there is no loose bodywork, for example. 

Different tyres and even tyre pressures can alter the road noise experienced, too.
